Research peptides ship as a lyophilized (freeze-dried) powder because peptides in aqueous solution degrade in weeks to months, while the same material as dry powder is stable for years when stored cold. Lyophilization is not packaging convenience — it is the step that makes a defined shelf life possible at all.
What lyophilization does
Freeze-drying removes water by sublimation: the solution is frozen, then held under vacuum so ice passes directly from solid to vapour without an intervening liquid phase. A secondary drying stage removes residual bound water. What remains is a porous solid cake — the visible puck or powder at the bottom of a vial.
The three stages, in order:
- Freezing. The solution is cooled below its eutectic point. Ice crystal structure formed here determines the porosity of the final cake.
- Primary drying. Under vacuum, with gentle heat, the ice sublimes. This removes the bulk of the water and is the longest stage.
- Secondary drying. Temperature is raised to drive off water still bound to the peptide, usually to a residual moisture content of a few percent or less.
Why water is the problem
Almost every degradation pathway available to a peptide requires water, mobility, or both:

Remove the water and molecular mobility collapses. Reaction rates that are meaningful over weeks in solution become negligible over years in a dry, cold, sealed vial.
Practical consequences for handling
- Do not open a cold vial. Ambient moisture condenses onto cold surfaces. Let the sealed vial reach room temperature first — the powder is hygroscopic and water introduced at this point starts the clock early.
- The cake may not look like much. 100 mg of lyophilized peptide occupies very little volume, and after transit the cake may be a thin film or loose powder rather than a neat puck. Cake appearance is not a purity indicator.
- Add solvent gently. Direct the stream against the vial wall, not onto the cake, and swirl rather than shake. Foaming denatures peptide at the air-liquid interface.
- Once reconstituted, the shelf life changes entirely. Dry-powder stability figures no longer apply. See storage and stability.
Why pre-mixed peptide is a warning sign
A supplier shipping peptide already in solution has given up the stability advantage that lyophilization exists to provide, and has done so before the material even leaves the warehouse. Transit time, transit temperature and time on the shelf all now count against a much shorter shelf life. There are legitimate pre-formulated products in pharmaceutical contexts, manufactured with stabilising excipients and cold-chain distribution. A research compound arriving pre-mixed in a plain vial is not that.
Room-temperature shipping is not a contradiction
Lyophilized peptides tolerate ambient temperature for the duration of normal transit. The dry state is what makes this acceptable — there is no aqueous chemistry to accelerate.
