Peptide degradation is quiet. There is rarely an obvious moment where material visibly goes bad — it simply loses potency while looking identical in the vial. Storage is therefore one of the few variables in research work that is entirely within your control, and one of the most commonly neglected.
Dry powder versus solution
The single most important distinction in peptide storage is whether the compound is still lyophilized or has been reconstituted.
As a dry, sealed, lyophilized powder, a peptide is remarkably stable. Kept at minus 20 degrees Celsius and protected from moisture and light, many compounds remain viable for years. The freeze-drying process removes the water that drives most degradation pathways.
Once reconstituted, that stability collapses. In solution, a peptide is typically stable for a matter of weeks under refrigeration rather than years. Water enables hydrolysis, oxidation and aggregation — the reactions that break peptide bonds and cause chains to clump together.
The practical consequence is straightforward: reconstitute only what you intend to use within a reasonable window, and keep the rest dry and frozen.
Temperature guidelines
Lyophilized, long-term storage: minus 20 degrees Celsius or colder. A standard freezer is adequate. Minus 80 extends stability further but is rarely necessary for typical timelines.
Lyophilized, short-term: refrigeration at 2 to 8 degrees Celsius is acceptable for weeks. Room temperature is acceptable for the short period of shipping, which is why peptides can be posted without dry ice, but it is not a storage condition.
Reconstituted solution: 2 to 8 degrees Celsius, refrigerated, always. Never at room temperature.
The freeze-thaw problem
Every freeze-thaw cycle damages peptide material. Ice crystal formation physically disrupts molecular structure, and the concentration changes that occur as water freezes out promote aggregation.
This is the most common avoidable error in peptide handling. A researcher freezes a reconstituted vial, thaws it for use, refreezes the remainder, and repeats — losing material each time while assuming the freezer is preserving it.
If a solution must be frozen, divide it into single-use aliquots first. Each portion is then thawed exactly once. The few minutes this takes preserves more material than any other single habit.
Light and oxygen
Several peptides are photosensitive, particularly those containing tryptophan, tyrosine or phenylalanine. Amber vials help; a closed box in the refrigerator works just as well and costs nothing.
Oxidation affects peptides containing methionine or cysteine. Keeping vials properly sealed and minimising the time they spend open limits exposure.
Moisture is the quiet enemy
Lyophilized peptide is hygroscopic — it pulls water from the air. This is why a cold vial should be allowed to reach room temperature before opening. Opening a cold vial in a warm room causes condensation to form inside it, introducing exactly the water that freeze-drying removed.
A practical routine
Store lyophilized vials in a freezer, in a sealed container, away from light. Before opening, let the vial warm to room temperature for 20 to 30 minutes. Reconstitute only what you need. Label every vial with compound, concentration and date. Refrigerate solutions and keep them dark. Aliquot before freezing, never after.
Storage cannot fix purity
All of this preserves what you started with. None of it improves it. If a vial contains less of the labelled compound than claimed, careful cold storage simply maintains that shortfall.
